Breast cancer is the most commonly diagnosed cancer in women globally, with approximately 310,000 new cases annually in the United States. Treatment is driven by three core molecular subtypes — HR+/HER2−, HER2+, and triple-negative (TNBC) — which determine systemic therapy sequencing across all disease stages. CDK4/6 inhibitors combined with endocrine therapy have transformed outcomes in metastatic HR+ disease, while the emergence of antibody-drug conjugates (T-DXd, sacituzumab govitecan) and PARP inhibitors for BRCA-mutated tumors has expanded late-line options substantially.
