Melanoma is the most lethal form of skin cancer, arising from melanocytes. Approximately 50% of cutaneous melanomas harbor an activating BRAF V600 mutation, making BRAF + MEK inhibitor combinations (dabrafenib/trametinib, vemurafenib/cobimetinib, encorafenib/binimetinib) a cornerstone of treatment for BRAF-mutant advanced disease. PD-1-based checkpoint immunotherapy (pembrolizumab, nivolumab ± ipilimumab) is the preferred approach for BRAF wild-type disease and is also approved for BRAF-mutant tumors in the adjuvant and metastatic settings.
