Multiple myeloma is a clonal plasma-cell malignancy accounting for approximately 10% of all hematologic malignancies. Treatment has been transformed by proteasome inhibitors, immunomodulatory drugs (IMiDs), CD38-directed monoclonal antibodies (daratumumab, isatuximab), and increasingly, BCMA-targeted bispecific antibodies and CAR-T cell therapies. Cytogenetic risk stratification — particularly del(17p), t(4;14), t(14;16), and gain(1q21) — guides therapy selection and transplant eligibility assessment, while FISH and next-generation sequencing define high-risk vs. standard-risk disease.
