Mechanism of Action

Bifunctional mechlorethamine derivative containing a purine-like benzimidazole ring. Mechanism is dual: alkylating agent activity (DNA cross-linking via nitrogen mustard moiety) plus antimetabolite activity (purine analog component disrupts DNA repair). This dual mechanism may explain activity in tumors resistant to classical alkylating agents.

Bendamustine-rituximab (BR) has demonstrated superiority to R-CHOP for indolent NHL (BRIGHT and StiL trials) with a more favorable toxicity profile and less alopecia, making it a preferred first-line regimen for follicular and marginal zone lymphoma.
