Mechanism of Action

Recombinant yeast-derived granulocyte-macrophage colony-stimulating factor (GM-CSF). Binds the GM-CSF receptor (GMR), a heterodimeric cytokine receptor expressed on myeloid progenitor cells, granulocytes, monocytes, macrophages, dendritic cells, and endothelial cells, activating JAK2/STAT5, PI3K/AKT, and MAPK signaling. Sargramostim differs from filgrastim/pegfilgrastim in stimulating the monocyte-macrophage-dendritic cell axis in addition to granulocytes — making it uniquely suited for situations requiring broad myeloid reconstitution (certain transplant settings) and for its immune adjuvant properties (investigated in cancer vaccines and immunotherapy combinations).
