Mechanism of Action

Oral imidazotetrazine prodrug that spontaneously converts to MTIC (methyltriazen-1-yl-imidazole-4-carboxamide) at physiological pH. MTIC methylates guanine at the O6 position, causing DNA mismatch, strand breaks, and apoptosis. Unlike streptozocin, temozolomide does not rely on GLUT2-mediated selective uptake into islet-derived cells — it distributes broadly to all tissues with good oral bioavailability and CNS penetration.
