BEACON CRC — Encorafenib, Binimetinib, and Cetuximab in BRAF V600E-Mutant Metastatic Colorectal Cancer (BEACON CRC)
Phase 3 · 2019 · Colorectal Cancer
Design
Randomized trial of encorafenib plus cetuximab (doublet) or encorafenib, binimetinib, and cetuximab (triplet) versus investigator's choice of cetuximab plus chemotherapy.
Population
BRAF V600E-mutant metastatic colorectal cancer after one or two prior regimens.
Primary Endpoint
overall survival and objective response rate (triplet vs control)
Key Result
Encorafenib plus cetuximab improved OS to 8.4 vs 5.4 months (HR 0.60) with a manageable toxicity profile; the doublet became the preferred regimen.
Guideline Impact
NCCN category 1 encorafenib plus cetuximab for previously treated BRAF V600E-mutant metastatic colorectal cancer
