ECOG 3200 — Bevacizumab in Combination With Oxaliplatin, Fluorouracil, and Leucovorin (FOLFOX4) for Previously Treated Metastatic Colorectal Cancer: Results From the Eastern Cooperative Oncology Group Study E3200

Phase 3 · 2007 · Colorectal Cancer

Design

Randomized phase III trial comparing FOLFOX4 plus bevacizumab, FOLFOX4 alone, and bevacizumab alone in previously treated patients.

Population

829 patients with metastatic colorectal cancer previously treated with a fluoropyrimidine and irinotecan (second-line setting).

Primary Endpoint

Overall survival

Key Result

Adding bevacizumab to FOLFOX4 improved median overall survival to 12.9 months versus 10.8 months for FOLFOX4 alone (HR 0.75, P=.0011) and improved progression-free survival (7.3 vs 4.7 months).

Guideline Impact

Supported bevacizumab plus FOLFOX in second-line metastatic colorectal cancer guidelines.
