JCOG 9907 — A Randomized Controlled Trial of Chemotherapy With 5-FU and Cisplatin Before and After Surgery for Stage II/III Squamous Cell Carcinoma of the Thoracic Esophagus (JCOG9907)
Phase III · 2012 · Esophageal Cancer
Design
Randomized phase III trial comparing two courses of cisplatin plus 5-fluorouracil given postoperatively (surgery first) versus preoperatively (chemotherapy first) in resectable thoracic esophageal squamous cell carcinoma.
Population
Patients with clinical stage II or III (excluding T4) squamous cell carcinoma of the thoracic esophagus.
Primary Endpoint
Progression-free survival
Key Result
Preoperative chemotherapy was superior, with 5-year overall survival of 55% versus 43% for postoperative chemotherapy (hazard ratio 0.73; 95% CI, 0.54-0.99; P = .04).
Guideline Impact
Made preoperative CF chemotherapy the Japanese standard for resectable stage II/III esophageal SCC
