Phase 3 · 2010 · Oropharyngeal Squamous Cell Carcinoma

Design

Randomized trial comparing accelerated-fractionation versus standard-fractionation radiotherapy, both with concurrent cisplatin.

Population

Stage III-IV squamous cell carcinoma of the head and neck, with prospectively collected HPV/p16 tumor status.

Primary Endpoint

overall survival (OS)

Key Result

3-year OS 82.4% for HPV-positive vs 57.1% for HPV-negative oropharyngeal cancer; HPV-positive status reduced death risk by 58%.

Guideline Impact

Established HPV/p16 testing and prognostic staging for oropharyngeal cancer
