RTOG 9111 — Concurrent Chemoradiotherapy Versus Induction Chemotherapy or Radiation Alone for Larynx Preservation

Phase 3 · 2003 · Head and Neck Cancer, Head and Neck Squamous Cell Carcinoma

Design

Randomized concurrent cisplatin-radiotherapy vs induction cisplatin/fluorouracil followed by radiation vs radiation alone.

Population

Locally advanced laryngeal squamous cell carcinoma otherwise requiring total laryngectomy.

Primary Endpoint

laryngectomy-free survival / larynx preservation

Key Result

Concurrent chemoradiotherapy gave the best larynx-preservation and locoregional control; induction chemotherapy and radiation alone were inferior for organ preservation.

Guideline Impact

Foundational evidence for organ-preserving chemoradiation in laryngeal cancer
