SWOG-S1826 — S1826: A Phase 3 Randomized Study of Nivolumab plus AVD versus Brentuximab Vedotin plus AVD in Newly Diagnosed Advanced-Stage Classic Hodgkin Lymphoma

Phase 3 · 2023 · Hodgkin Lymphoma

Design

Randomized phase 3 trial comparing nivolumab plus AVD (doxorubicin, vinblastine, dacarbazine) versus brentuximab vedotin plus AVD for six cycles.

Population

Patients 12 years or older with newly diagnosed advanced-stage (stage III or IV) classic Hodgkin lymphoma.

Primary Endpoint

Progression-free survival

Key Result

At a median follow-up of 2.1 years, 2-year progression-free survival was 92% with nivolumab-AVD versus 83% with brentuximab vedotin-AVD (HR for progression or death 0.45, 95% CI 0.30-0.65; P<0.001), with lower toxicity.

Guideline Impact

Adopted into NCCN guidelines as a preferred first-line regimen for advanced-stage classic Hodgkin lymphoma
