CALGB 8461 — Cytogenetic Studies in Acute Leukemia and Multiple Myeloma (CALGB 8461 cytogenetics companion program)
Correlative / companion study · Acute Myeloid Leukemia, Acute Leukemia, Multiple Myeloma
Population
Adults with newly diagnosed acute leukemia (including de novo AML) or multiple myeloma enrolled on companion CALGB/Alliance treatment studies.
Primary Endpoint
Association of pretreatment cytogenetic abnormalities with induction success, relapse, and overall survival
Rationale
Pretreatment cytogenetics are among the strongest prognostic markers in acute leukemia, but standardized, prospectively collected data across large cooperative-group cohorts are needed to define and continually refine risk categories. This companion program supplies that correlative backbone.
What It Could Change
Continues to underpin cytogenetic risk stratification in AML — guiding remission expectations and selection for allogeneic transplant — and refines molecular risk groups as new abnormalities are characterized.
