Untreated, HER2-negative, CLDN18.2-positive locally advanced or metastatic gastric or gastroesophageal junction adenocarcinoma.

Rationale
Roughly a third of gastric/GEJ adenocarcinomas express CLDN18.2. GLOW tests whether adding the anti-CLDN18.2 antibody zolbetuximab to first-line CAPOX chemotherapy improves outcomes in this biomarker-selected population.
What It Could Change
Established zolbetuximab plus CAPOX as a first-line option for CLDN18.2-positive, HER2-negative gastric/GEJ adenocarcinoma, adding a biomarker-directed backbone to chemotherapy.
