PSMAfore — 177Lu-PSMA-617 radioligand therapy versus a switch of androgen-receptor pathway inhibitor in taxane-naïve metastatic castration-resistant prostate cancer (PSMAfore)

Phase 3 · Prostate Cancer

Population

PSMA-positive, taxane-naïve metastatic castration-resistant prostate cancer that has progressed on one prior androgen-receptor pathway inhibitor.

Primary Endpoint

Radiographic progression-free survival

Rationale

177Lu-PSMA-617 is established after both an androgen-receptor pathway inhibitor and taxane chemotherapy. PSMAfore tests whether moving radioligand therapy earlier — before chemotherapy — beats simply switching to a second hormonal agent.

What It Could Change

Could bring PSMA-targeted radioligand therapy into the pre-chemotherapy setting for castration-resistant prostate cancer.
