Mechanism of Action
Conditionally-activated ("masked", POWERbody/SAFEbody-engineered) agonist antibody targeting CD137 (4-1BB), a potent T-cell costimulatory receptor. A protease-cleavable mask blocks binding in normal tissue and is selectively removed by tumor-associated proteases in the tumor microenvironment, restoring CD137 agonism locally. This design plus Fc engineering aims to deliver strong 4-1BB costimulation of tumor-reactive T cells while avoiding the systemic T-cell activation and hepatotoxicity that limited earlier 4-1BB agonists.
