Mechanism of Action
Fully synthetic third-generation 9-aminoanthracycline and a potent topoisomerase II inhibitor. Amrubicin, and its more active C-13 hydroxy metabolite amrubicinol, stabilize the topoisomerase II-DNA cleavable complex, producing DNA double-strand breaks and apoptosis. Unlike classic anthracyclines (doxorubicin, epirubicin), its cytotoxicity is driven predominantly by topoisomerase II poisoning rather than DNA intercalation and free-radical generation, which is associated with a substantially lower risk of cumulative cardiotoxicity.
