Mechanism of Action
Arlocabtagene autoleucel is an autologous chimeric antigen receptor (CAR) T-cell therapy directed against G-protein-coupled receptor class C group 5 member D (GPRC5D), an orphan receptor highly expressed on malignant plasma cells with limited normal-tissue distribution. Patient T cells are engineered ex vivo to express the anti-GPRC5D CAR, then reinfused after lymphodepleting chemotherapy. Upon binding GPRC5D on myeloma cells, the CAR triggers T-cell activation, cytokine release, and cytotoxic killing.
