Mechanism of Action
Selective small-molecule inhibitor of DNA polymerase theta (Pol-theta/POLQ), the key effector of microhomology-mediated end joining. In HR-deficient tumors, cells become dependent on Pol-theta for double-strand break repair, so inhibition is synthetically lethal and enhances PARP inhibitor activity, potentially overcoming PARPi resistance.
