Mechanism of Action
AZD0901 (CMG901) is a Claudin18.2-targeting antibody-drug conjugate consisting of a humanized anti-CLDN18.2 monoclonal antibody linked to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline linker. It binds CLDN18.2 selectively expressed on gastric and GEJ adenocarcinoma cells, undergoes receptor-mediated internalization, and releases MMAE intracellularly to inhibit tubulin polymerization and trigger G2/M arrest and apoptosis.
