Mechanism of Action
First-in-class CD8-guided T-cell engager (TCE) targeting glypican-3 (GPC3), an oncofetal cell-surface proteoglycan highly expressed on hepatocellular carcinoma and other solid tumors but largely absent from normal adult tissue. Unlike conventional GPC3 × CD3 bispecifics, AZD9793 incorporates a CD8-targeting arm in addition to its GPC3- and CD3-binding domains, preferentially redirecting and activating cytotoxic CD8+ T cells at the tumor while limiting CD4+-driven cytokine release.
