Mechanism of Action

Azercabtagene zapreleucel (azer-cel) is an allogeneic, off-the-shelf CD19-directed CAR T-cell therapy manufactured from healthy-donor T cells using ARCUS genome-editing to disrupt the endogenous T-cell receptor and reduce graft-versus-host risk. The engineered cells express an anti-CD19 CAR that recognizes CD19 on malignant and normal B cells, triggering T-cell activation and cytolytic killing. Because it is allogeneic and cryopreserved, it is available without patient-specific manufacturing delay. Lymphodepletion, sometimes with an added immunomodulatory component, supports CAR-T persistence.
