Mechanism of Action
Sevabertinib (BAY 2927088) is an oral, reversible, potent tyrosine kinase inhibitor with activity against mutant HER2 (ERBB2), including HER2 exon 20 insertion mutations, as well as mutant EGFR. It selectively inhibits aberrant HER2/EGFR kinase signaling while relatively sparing wild-type EGFR, an approach intended to preserve efficacy against exon 20 insertion drivers while reducing the wild-type EGFR-mediated toxicities (rash, diarrhea) typical of non-selective EGFR TKIs.
Investigational Indications
- HER2 (ERBB2)-mutant non-small cell lung cancer (NSCLC), including HER2 exon 20 insertion mutations
- EGFR-mutant NSCLC (including exon 20 insertions)
- Other HER2-mutant advanced solid tumors
