First-in-class dual ON/OFF KRAS G12C inhibitor that covalently binds and inhibits both the active (GTP-bound, ON) and inactive (GDP-bound, OFF) states of KRAS G12C, aiming for deeper pathway suppression than OFF-state-only inhibitors.
