Bemarituzumab — Investigational Agent

Mechanism of Action
Humanized, afucosylated IgG1 monoclonal antibody targeting the FGFR2b splice isoform. It blocks FGF ligand binding to FGFR2b, inhibiting downstream oncogenic signaling, and its afucosylated Fc enhances antibody-dependent cellular cytotoxicity against FGFR2b-overexpressing tumor cells. It selectively acts on tumors overexpressing FGFR2b (assessed by immunohistochemistry) and/or with FGFR2 amplification.
Investigational Indications
- First-line FGFR2b-overexpressing advanced gastric / gastroesophageal junction adenocarcinoma (with chemotherapy +/- nivolumab)
