Mechanism of Action
BI 1823911 is an oral, covalent inhibitor of KRAS G12C that binds the mutant cysteine-12 residue and traps KRAS G12C in its inactive GDP-bound state, blocking downstream RAS-MAPK signaling. It is being developed both as monotherapy and in rational combinations, notably with the SOS1 inhibitor BI 1701963, aiming to deepen and prolong pathway suppression and delay adaptive resistance.
