Mechanism of Action

Bomedemstat is an orally administered, irreversible inhibitor of lysine-specific demethylase 1 (LSD1/KDM1A), a flavin-dependent enzyme that regulates the epigenetic transcriptional program governing hematopoietic stem and progenitor cell differentiation. In myeloproliferative neoplasms, LSD1 is required for the survival and self-renewal of malignant megakaryocyte and progenitor populations. Inhibition promotes differentiation and apoptosis of the neoplastic clone, reducing pathologic megakaryopoiesis and platelet overproduction.
