Mechanism of Action
Camizestrant is a potent, oral, next-generation selective estrogen receptor degrader (SERD) and pure ER antagonist. It binds the estrogen receptor (ER), blocks estrogen-driven transcription, and promotes ER degradation. It retains full antagonist activity against clinically important ESR1 ligand-binding-domain mutations (e.g., Y537S, D538G) that confer resistance to aromatase inhibitors, providing continuous ER pathway suppression without the intramuscular delivery limitations of fulvestrant.
Investigational Indications
- HR+/HER2- advanced/metastatic breast cancer with emergent ESR1 mutation (ctDNA-guided switching)
- HR+/HER2- early and advanced breast cancer (combination with CDK4/6 inhibitors)
