Mechanism of Action
Fully human agonistic IgG2 monoclonal antibody targeting CD40, a costimulatory receptor on antigen-presenting cells (dendritic cells, macrophages, B cells). CD40 agonism licenses dendritic cells to prime tumor-specific CD8+ T cells and repolarizes tumor-associated macrophages toward a tumoricidal phenotype. CDX-1140 was engineered for potent agonism with a wide therapeutic window, allowing systemic dosing without the dose-limiting hepatotoxicity that constrained earlier CD40 agonists.
