Mechanism of Action
Cemsidomide is an orally bioavailable cereblon-modulating agent (CELMoD) and a molecular glue degrader that recruits the neosubstrate transcription factors IKZF1 (Ikaros) and IKZF3 (Aiolos) to the cereblon E3 ubiquitin ligase complex. This drives their polyubiquitination and proteasomal degradation with high potency and rapid kinetics. Loss of Ikaros and Aiolos suppresses transcriptional survival programs in malignant plasma cells and B-lymphocytes and augments T-cell and NK-cell immune activity.
