Mechanism of Action

Fully human IgG1 monoclonal antibody targeting VISTA (V-domain Ig suppressor of T-cell activation; also called PD-1H), a negative checkpoint regulator highly expressed on myeloid-derived suppressor cells, tumor-associated macrophages, and regulatory T cells. By blocking VISTA-mediated inhibitory signaling, CI-8993 relieves suppression of T-cell activation and reprograms the myeloid compartment of the tumor microenvironment toward a pro-inflammatory state, representing a distinct checkpoint axis from PD-1/PD-L1 and CTLA-4.
