Mechanism of Action
Cretostimogene grenadenorepvec is an intravesical oncolytic serotype-5 adenovirus engineered with an E2F-1 promoter driving E1A expression, restricting viral replication to Rb-pathway-defective tumor cells (nearly all bladder cancers). Selective replication lyses malignant urothelial cells, and the virus also encodes GM-CSF to recruit and activate dendritic cells and promote a systemic anti-tumor immune response (immunogenic cell death).
