Mechanism of Action
Pan-RAS RAS(ON) inhibitor that binds the switch-II pocket of GTP-bound (active) RAS, locking it in its active conformation and preventing downstream effector engagement. Unlike sotorasib and adagrasib, which covalently target KRAS G12C in the GDP-bound (inactive) state, daraxonrasib targets all GTP-loaded oncogenic RAS isoforms (KRAS G12X, G13D, NRAS, HRAS) without isoform restriction.
