Disitamab vedotin — Investigational Agent
Antibody-Drug Conjugate · Phase 3 · RemeGen / Pfizer (ex-Seagen)
Mechanism of Action
Disitamab vedotin is a HER2-directed antibody-drug conjugate combining a humanized anti-HER2 monoclonal antibody (hertuzumab) linked via a cleavable linker to monomethyl auristatin E (MMAE). After binding HER2 and internalization, MMAE is released, disrupting microtubules and triggering apoptosis. Activity extends to HER2-low as well as HER2-overexpressing tumors, with bystander killing of neighboring cells.
Investigational Indications
- HER2-expressing locally advanced or metastatic urothelial carcinoma
- HER2-overexpressing advanced gastric / gastroesophageal junction cancer
- Other HER2-expressing solid tumors
