Mechanism of Action

Domvanalimab is an Fc-silent, humanized IgG1 monoclonal antibody targeting the TIGIT receptor. Unlike Fc-enabled TIGIT inhibitors, domvanalimab is designed to block the TIGIT/PVR interaction without depleting TIGIT-expressing cells (like peripheral Tregs or effector T-cells) via ADCC. This "Fc-silent" approach is hypothesized to maximize the availability of effector T-cells and reduce potential toxicities. It is intended to restore T-cell and NK cell activation in the tumor microenvironment.
