Mechanism of Action
First-in-class imipridone that acts as a selective antagonist of dopamine receptor DRD2 and a potent agonist of the mitochondrial protease ClpP. ClpP hyperactivation triggers degradation of respiratory chain subunits, collapsing oxidative phosphorylation and inducing the integrated stress response (ATF4/CHOP) preferentially in H3 K27M-mutant diffuse midline glioma cells.
