Mechanism of Action
Selective small-molecule inhibitor of the Werner syndrome RecQ helicase (WRN). Microsatellite-instability-high (MSI-H)/mismatch-repair-deficient tumors depend on WRN to resolve secondary DNA structures at expanded (TA)n repeats; WRN inhibition triggers catastrophic double-strand breaks selectively in MSI-H cells, a novel synthetic-lethal vulnerability.
