Mechanism of Action
Orally bioavailable, potent and selective inhibitor of ataxia telangiectasia and Rad3-related (ATR) kinase. Blocks ATR-mediated DNA damage response signaling at stalled replication forks, driving replication catastrophe and synthetic lethality in tumors with high replication stress or defects in ATM/DNA-repair pathways, and potentiates DNA-damaging chemotherapy.
Investigational Indications
- DNA damage response-deficient advanced solid tumors (e.g., ATM-altered)
- Platinum-resistant ovarian cancer (with chemotherapy)
- Advanced pancreatic and other solid tumors
- Replication-stress-high gynecologic cancers
