Mechanism of Action
Oral small-molecule inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4), a key node in Toll-like receptor and IL-1R innate-immune signaling upstream of NF-kB. IRAK4 hyperactivation (including the oncogenic IRAK4-long isoform driven by U2AF1/SF3B1 spliceosome mutations) sustains survival signaling in myeloid malignancies and MYD88-mutant lymphomas. Emavusertib also inhibits FLT3, giving activity in FLT3-mutant AML.

- Higher-risk myelodysplastic syndrome (MDS)
- Relapsed/refractory non-Hodgkin lymphoma (primary CNS lymphoma; MYD88-mutant)
