Mechanism of Action

A high-affinity CD47-blocking fusion protein: an engineered SIRPα variant domain linked to an inactivated (Fc-dead) human IgG1 Fc. By binding CD47 with picomolar affinity while carrying a Fc engineered to minimize red-cell binding and hematologic toxicity, evorpacept blocks the CD47–SIRPα "don't eat me" signal to enhance macrophage phagocytosis — with substantially less on-target anemia than antibody-based anti-CD47 agents. Designed to be combined with tumor-targeting antibodies (rituximab, trastuzumab, cetuximab) and other standards of care.
