Mechanism of Action
Garsorasib is an orally administered, covalent, selective inhibitor of KRAS G12C. It binds irreversibly to the mutant cysteine-12 in the switch-II pocket, locking KRAS G12C in the inactive GDP-bound conformation and blocking downstream RAF-MEK-ERK and PI3K-AKT signaling. It is highly selective for the G12C mutant over wild-type KRAS and other RAS isoforms.
