Mechanism of Action
Givastomig is a Claudin18.2 x 4-1BB (CD137) bispecific antibody that conditionally activates T-cell costimulation only in the tumor microenvironment. The CLDN18.2 arm anchors the molecule to gastric, GEJ, and pancreatic tumor cells, and this target engagement is required to cluster and activate 4-1BB on tumor-infiltrating T cells. This tumor-localized 4-1BB agonism enhances T-cell proliferation, survival, and cytotoxicity while avoiding the systemic hepatotoxicity seen with unconditional 4-1BB agonists.
