Mechanism of Action
Glecirasib is a potent and highly selective small-molecule inhibitor of the KRAS G12C mutant protein. It works by binding to the switch-II pocket (P2) of the KRAS G12C protein in its inactive (GDP-bound) state. This covalent lock prevents the protein from transitioning to the active (GTP-bound) state, thereby halting the constitutive activation of downstream signaling pathways such as RAF/MEK/ERK. The inhibition of these pathways leads to decreased tumor cell proliferation and induction of apoptosis specifically in KRAS G12C-mutant tumors.
