Mechanism of Action
Golcadomide is a potent, small molecule Cereblon E3 Ligase Modulator (CELMoD) optimized for the treatment of non-Hodgkin lymphomas. Like other CELMoDs, it binds to cereblon, facilitating the recruitment and proteasomal degradation of the lymphoid transcription factors Ikaros and Aiolos. In lymphoma cells, this degradation results in cell cycle arrest and apoptosis, particularly in Germinal Center B-cell (GCB) and Activated B-cell (ABC) subtypes of DLBCL.
