Mechanism of Action
Gotistobart is a next-generation, pH-sensitive anti-CTLA-4 monoclonal antibody engineered to preserve CTLA-4 recycling. Conventional anti-CTLA-4 antibodies bind CTLA-4 in acidic endosomes and target it for lysosomal degradation, depleting the receptor and reducing regulatory T-cell function systemically, driving immune-related toxicity. Gotistobart dissociates from CTLA-4 at low endosomal pH, allowing CTLA-4 to recycle to the cell surface while still blocking B7 costimulation and enabling preferential depletion of intratumoral regulatory T cells via ADCC.
