Mechanism of Action

First-in-class allosteric, non-covalent WRN helicase inhibitor that binds between the helicase D1/D2 domains, locking WRN in an inactive DNA-bound state and triggering ATM/DNA-PK-dependent DNA damage selectively in MSI-H/dMMR cells.
