Mechanism of Action
A first-in-class monoclonal antibody that selectively binds the mutant calreticulin (CALR) neoepitope on the cell surface, blocking mutCALR-driven activation of the thrombopoietin receptor MPL. This disrupts constitutive JAK-STAT signaling that drives clonal proliferation in CALR-mutant myeloproliferative neoplasms while sparing wild-type cells.
