Mechanism of Action
Autologous engineered T-cell therapy expressing an affinity-enhanced T-cell receptor (SPEAR T-cell) directed against a peptide from the NY-ESO-1 / LAGE-1a cancer-testis antigen presented by HLA-A*02. The patient's T cells are harvested, transduced ex vivo to express the NY-ESO-1–specific TCR, expanded, and reinfused after lymphodepleting chemotherapy, enabling recognition and killing of NY-ESO-1–expressing tumor cells. (Note: the assigned brief labeled this MAGE-A4 — that target belongs to afami-cel/Tecelra, which is FDA-approved; lete-cel is NY-ESO-1–directed.)
