Mechanism of Action
¹⁷⁷Lu-PSMA-617 is a radioligand therapy that couples the beta-emitting radionuclide lutetium-177 to a small molecule targeting prostate-specific membrane antigen (PSMA), a transmembrane glycoprotein highly overexpressed on prostate cancer cells. After binding PSMA, the complex is internalized and delivers cytotoxic beta radiation (~2 mm path length) causing double-strand DNA breaks and cell death while relatively sparing surrounding tissue.
