Mechanism of Action
Mezigdomide is a first-in-class Cereblon E3 Ligase Modulator (CELMoD) agent specifically optimized for maximal degradation of Ikaros (IKZF1) and Aiolos (IKZF3). It binds to the cereblon E3 ubiquitin ligase complex, inducing a conformational change that recruits these lymphoid transcription factors for polyubiquitination and subsequent proteasomal degradation. The rapid and deep degradation of Ikaros and Aiolos leads to direct apoptosis of malignant B-cells and potent stimulation of T-cells and NK-cells.
